The names R2-D2 and C-3PO are actually abbreviations that come from filmmaking jargon:
R2-D2 stands for "Reel 2, Dialogue 2" — it was originally a shorthand used by the film editor George Lucas heard on the set of American Graffiti to describe a piece of film. He liked the sound and used it for the droid’s name.
C-3PO stands for "Cypher 3 Protocol" — reflecting the droid’s role as a protocol and translation robot.
So, while not acronyms with deep in-universe meaning, their names came from technical film terms and were adapted into the Star Wars universe.
